DNA template is the DNA that is used for PCR which contains the region of the DNA fragment to be amplified.

Two primers are used to determine the start and end of the region to be amplified.
DNA templates can simply be crude extracted DNA, vector or plasmid DNA, even cDNA.
You can prepare DNA for PCR by mini-prep, midi-prep or maxi-prep if you are preparing plasmids from bacteria.
