Overview of EMSA Procedure

Function of the EMSA is that it is an assay which checks for the presence of protein-DNA binding.


Materials Needed to Perform DNA EMSA

DNA EMSA Electrophoresis

Loading solution

  • It is usually light sensitive so store in dark area (cover with aluminum foil)
  • stored at -20°C
  • comes in EMSA kits from Pierce (Light Shift Chemiluminescent Kit) and Invitrogen (manual, catalog number - E-33075)
  • Typical DNA gel loading buffer can also be used.

Running Buffer for Electrophoresis

0.5X TBE running buffer

  • 44.5 mM Tris base
  • 44.5 mM Boric acid
  • 1 mM EDTA (free acid)
  • pH usually around 7-8
  • You can make yourself or buy from a supplier such as Invitrogen (catalog number LC6675)
