Protocol for DNA fragment purification from agarose or acrylamide. For fragments from 200 bp to 10 kb the agarose purification is ideal. For smaller fragments (20 bp to 400 bp) the acrylamide purification is preferred. - [Read DNA Fragment Purification from Agarose or Acrylamide Protocol]
This is a rapid alkaline lysis miniprep method for isolating DNA from large PAC clones. It is a modification of a standard Qiagen-Tip method that uses no organic extractions or columns. The method works very well for doing analytical restriction digests of PAC clones and can be scaled up if necessary. - [Read DNA Isolation From BAC & PAC Clones Protocol]
Protocol for fungal DNA isolation. The key elements in this prep are (1) the use of young lyophilized mycelial mats....young mats (4 days growth for C. carbonum)...yield less contaminating carbohydrates and other misc. junk (2) lots of proteinase K in the extraction buffer to kill Dnases (final =0.3mg/ml). - [Read Fungal DNA Isolation Protocol]
Protocol for single strand DNA prep for sequencing. This protocol works well from a 5 ml starting culture or a 1 ml starting culture (adjust volume accordingly). - [Read Single Strand DNA Prep For Sequencing Protocol]
