Rabbit-derived corneal cells are cultured in the presence of test compounds, the toxicity of which are determined by their effect upon cell viability. A decrease in cell number, as measured by uptake of the dye Neutral Red, serves as an indicator of potential cytotoxicity. Accumulation of lipophilic substances, many of which may be environmental chemicals, affects the membrane lipid order and consequently affects the functions of these proteins. This protocol describes the electroporation of the BMH 81-17 mut S strain that is recommended for tranformation of the site directed mutagenesis of dsDNA (See Protocol on Site-Directed Mutagenesis on Double Stranded DNA). BMH 81-17 mut S are a mismatch repair defective (mut S) Escherichia coli strain. The probability that the two mutations will cosegregate during the first round of DNA replication is increased in this strain.
