Analysis of DNA Fragmentation Using the JAM Assay. By Shailaja Kasibhatla et al., The JAM assay is based on labeling nuclear DNA of cycling cells with [3H]thymidine and harvesting samples on glass fiber filters. Apoptosis will generate DNA fragments small enough to pass through the glass fiber filter, resulting in decreased radioactivity of the particular sample. Cell-mediated cytotoxicity or cell killing mediated by cytotoxic T lymphocytes (CTL) can also be measured by this technique. - [Read Analysis Of DNA Fragmentation Using The JAM Assay (Subscription Required)]

Cryogenic preservation (storage below -100°C) of cell cultures is widely used to maintain backups or reserves of cells without the associated effort and expense of feeding and caring for them. The success of the freezing process depends on four critical areas: Proper handling and gentle harvesting of the cultures; Correct use of the cryoprotective agent; A controlled rate of freezing; Storage under proper cryogenic conditions. - [Read Cryogenic Preservation and Storage of Animal Cells Protocol]

Protocol is the first in a set of three describing fluorescent mRNA differential display (FDD or FDDRT-PCR). The method begins with the harvesting of total RNA from the tissue-cultured cells of interest. For other starting materials, such as blood samples, please see Extraction and Purification of RNA from Blood Samples for Fluorescent mRNA Differential Display. - [Read Extraction and Purification of RNA from Tissue-Cultured Cells for Fluorescent mRNA Differential]

Stably transfected cells, generated in the first two stages of the procedure, are induced for expression of the target gene. After harvesting and lysis, the lysates are analyzed by SDS-PAGE and immunoblotting. - [Read Tetracycline as Regulator of Inducible Gene Expression III]
Protocol describes the trypsinization of cells in monolayer culture to facilitate subculture or harvesting. To avoid cross-contamination of cells, it is important for each cell line to be subcultured independently. No more than one cell line should be in the tissue culture hood at any one time. - [Read Trypsinization of Cells Grown in Monolayer Protocol]
