Step by Step Method for Purifying DNA Fragments from Agarose Gels


  1. First, excise your band of interest from the agarose-TAE gel
  2. Estimate the volume by weighing the gel slice on a scale.
  3. Dissolve the agarose gel slice in 3 volumes of NaI (either make your own or obtain from a Geneclean kit) at 55oC for 5 minutes or more until the gel dissolves.
  4. Add 1 ml of Wizard miniprep resin (Promega), 10 min. at room temperature.
  5. Add the DNA resin mixture to a syringe and pass through a minicolumn.
  6. Wash with 2 ml of Wizard wash buffer; spin 2 min in a microfuge.
  7. Elute with 70oC TE or water (50 ul), 2 min in a microfuge.
