Simple and Quick Step By Step Agarose Gel Electrophoresis Method

  1. To make a 1% agarose gel, dissolve 1 g of agarose in 100 ml of 1X TAE or TBE buffer (sometimes the gel electrophoresis requires only 0.5X buffer - see your manual for more information)
  2. Microwave with tissue in the top of the flask
  3. Careful agarose is burning hot when boiling!
  4. Cast the gel with the comb in place once a bit cooler.
  5. When the gel has solidified well (15-30 minutes +), remove the comb.
  6. Add 6X gel loading buffer to sample and load the samples into wells.
  7. Run the gel submerged in 1X TAE or TBE (30-60 min. at 100-150V)
  8. Stain the gel with ethidium bromide solution (10 µl per 100ml of buffer) for 10 - 15 min.
  9. View on a long wave UV transilluminator.
