Protocol for Lambda phage DNA - Recovering DNA from Large Maxi Preparations
Collect phage from SM dialysis
add EDTA to equal 20mM
add Proteinase K to equal 50 ug/ml.
add SDS to equal 0.5%
incubate at 65oC for 1 hr
extract with equal volume phenol
extract with equal ratio phenol/chloroform to total equal volume
extract with equal volume chloroform
dialyze against 1X T.E. overnight or longer
