| Hey all, I know you can't run Tris-HEPES gels with Tris-Glycine-SDS running buffer, but can you run Tris-Glycine gels with Tris-HEPES-SDS running buffer? Thanks!! |

| Precast gel from Invitrogen (Novex) is made with Tris-Glycine buffer, while gel from Thermo Scientific Pierce is produced with Tris-HEPES buffer, both for homogeneous and gradient electrophoresis. Both gels do not contain SDS that is incorporated in the run buffer. Run nonreducing buffers from you described Tris-HEPES-SDS (10x : 121 g/L Tris base, 238 g/L HEPES and 10/L g SDS at pH pH 8.0 ± 0.25) or Tris-Glycine-SDS (10x : 29 g/L Tris, 144 g/L Glycine and 10 g/L SDS at pH 8.3), used in 1x form, they is compatible with both gels but each buffer produces different migration patterns. |
