Question Gel breakage during transfering on electroblotter! HELP PLEASE



Hello,

I just would like to know, if the gel breakage ( it broke in a few places) during transferring onto electro blotter has any bad impact on proper protein's bands transfer onto membrane?

( I put all pieces back together again, and started transferring gel onto membrane. can anyone answer me, if that broken gel could be still used on electro blotter? ?) would I be still able to see the bands of interst after adding 2 nd antibody?
Please Help!

P.s I use semi-dry transfer method!

Yes, the protein bands will still transfer onto the membrane even if the gel has been broken at some areas. I use semi-dry transfer method and had my gel broken too. Needless to say, I panicked but I went ahead to piece the bits up properly (you must put the pieces back properly on the membrane) and went ahead with the protein transfer. It works.
