| Does anyone out there test for human IgE to allergen proteins? I am having problems with a patient serum sample that shows binding without the mouse anti-human IgE monoclonal. The Goat anti-mouse (alk phos conjugated) is binding the protein bands with her serum with or without the mouse anti-human IgE. The goat antibody is adsorbed for Ig to human IgG. I run a known Mt Cedar allergic patient that is known to be IgE mediated and it is binding only with the mouse anti-human IgE - not binding without it. This is the way it is supposed to work. The patient I am having the trouble with is suspected of Ige mediated allergy to Annatto seed. All the blots are nice and clean with no background. I have tried diluting the serum, the antibodies, heat treating the serum for IgM inactivation in case IgM is non-specifically binding - but the serum is always binding and coming up with bands. Has anyone ever had trouble like this that may be patient specific -like drug interferences or disease states that would cause this? Will be waiting for help... |
