Transglutaminase

I am having hard time doing WB for Anti-Human Keratinocyte Transglutaminase
it's 90KD protein.I have tried 7.5% and 10%gel. I do not get band ,though loading 80ug of protein.

I tried ----

1 hour transfer at100V
2hour transfer at 90v
3hour transfer at60v

I need help

I have no experience with Invitrogen bis- tris and tris-acetate precast gels. If anyone can guide me to use them for transglutaminase will it work out.
