| Hi all- I'm trying to generate a stable cell line using nucleofected C3H cells under G418 selection. Everything goes great until I do a limit dilution to select for single cells- at this point, the cells are all dying. I'm guessing it's due to the single cell not being able to properly condition the media. Any suggestions on how to fix this? I've thought of using a different media- maybe Ham's cloning, but I have no experience in this so I thought I'd ask. Thanks for any help! -Anne |
