Change Media With Transfection?
 
Hello,
is it a good idea to change the media when you transfect cells with dna and lipofectamine or fugene etc? If so, why is this done?

Hi,

It is important to change the media because proteins in SFB or the antibiotics may interfere with the lipofectamine, and enter trough the cells membrane along with your DNA, then the efficiency of transfection decrease. But 4 hr affter transfection you must change the medua again... otherwise your cells could die...

I recently used a transfection reagent called DNAFect LT, which showed excellent performance. There is no need to change medium and no medium change for transfection. You may have a try.

We do change media just before transfection, and if you change 24 h later this would increase your transfection efficiency.

Hi
another alternative is to use magnetofection. After 30 min of incubation of your transfection reagents with cells on a magnetic plate, you can wash your cells (the magnet keeps the transfecting particules at the cell surface while you remove the medium) with fresh medium.
with magnetofection, washing is just an option since the doses are less important thant with other lipid reagents

Yeah, it is better to keep your happy and to increase the transfection efficiency. Therefore you must to change the media 4 hrs post transfection with lipofectamine.
