Default mice Mesenchymal Stem Cells (MSCs)



I am working on Mice MSCs and getting problem in trypsinization of cells to passage them. It is very hard to trypsinize and detach mice MSCs and it take very long time and even then don't detach fully. This is not the case with rat or human MSCs. Do anybody experience the same problem or is there is somebody who have solution for this problem.

I think the problem is caused due to haematopoetic stem cells (HSC) contamination that also get adhere to coated plastic surfaces and they are usually resistant to trypsin treatment. HSC gradually die in MSC medium and after few passaging this problem is removed.
So we need to adopt a special procedure to isolate miceMSC
A recently published paper describe this problem and give a better way to isolate mice MSC
