Exclamation Low Homologous Recombination Rates Stem Cells

Hello, i am new to the gene targeting gane and to this amazing forum also

I am conducting gene targeting experiments using mouse embyronic ES stem cells, G418 positive and DT negative selection.

My problem is the rate of homologous recombinant clones which actually survive is quite low ~1:500


Anyone have suggestions to increase this frequency?
