| Insulin-producing cells derived from human foreskin cells In vitro The success of insulin-producing cell transplantation in type 1 diabetes indicates that cell replacement therapy of this severe disease is achievable. However, the severe lack of donor cells has increased the demand for alternative cell sources. Here, we reported a novel stem cells from human foreskin subcutaneous tissue, which expressed pluripotent markers Oct 4, Sox 2, Rex 1, Nanog stably in the medium without b FGF and LIF supplemented. In the serum-free system, foreskin cells were induced into insulin-producing cells by three stage treatment. Activin A and All-trans retinoic acid (RA) were used to induce foreskin cells into pancreatic differentiation, as detected by the expression of endoderm marker Sox17 and the early pancreatic transcription factors Pdx1. After maturation with b FGF and nicotinamide, the insulin-producing cells expressed C-peptide, Insulin, Glut2, Glucokinase and Sur 1, and the percent of Glut 2 positive cells was about 58.31%. The secretion of insulin by these cells corresponding to 5 m M and 25 m M glucose levels were 4.17 μU/mg and 9.30 μU/mg respectively. So foreskin cells provide a novel stem cell source used for the treatment of type I diabetes in the future. |
