Default Detection of Akt1, GSK-3b and b-catenin in PBMCs

Hi, everyone.
Does anybody used western blot for detection of Akt1, GSK-3b and b-catenin (and their phospho forms) in PBMCs? Are there any special features or pitfalls? Maybe someone can advise cell concentration in lysis buffer and ammount of protein loading suitable for the best signal.

Thanks.
