Default Shift of phospho/phospho antibodies

Hi everybody,

does someone know a good way to show a phospho-shift in the signal transduction? I mean shall I treat the lysate with a phosphatase and load on to the SDS-PAGE (enzyme-treated vs untreated) or is it sufficiant to incubate with phospho specific abs? Additionally I have just found antibody against tyr. What about serine and threonine? Does anybody know good antibodies?
Thanks for any comments!!
