| dear all, can somebody tell me is it necessary to pre run the gel in running buffer before loading the sample.if it is to be done what is its purpose. |

| Hello Prachigptgene, No it is not necessary to pre-run SDS-PAGE gels prior to running the samples. Simply fill the inner and outer buffer reservoirs, load your samples in the lanes, and run. |

| Hi there, Pre-running of an acrylamide gel was necessary in DNA sequencing gels to get the temperature up and reduce the smiling fronts of bands ("smiley bands"). It is not necessary in discontinuous gel (stacking & resolving) SDS-PAGE gels |
