| Dear all, The usual protocols for SDS-PAGE gel preparation specify that the gel must be run in the same day it is prepared. Is it possible to prepare the gel (casting and pouring the solution into glass slides) and run it with the samples the day after? Many thanks. |

| I've never done that with SDS-PA gels, but when I worked with non-denaturing PA gels (only PA, without SDS) for DNA electrophoresis, we used to do what you say (casting the gels and storing them at 4ºC) and they worked fine. We stored them in the fridge wrapped with plastic bags. If you try this with SDS-PA gels, please let me know your results. Thanks |

| In the good ole days, we poured gels in the glass plates, wrapped them with plastic wrap, placed them in the cold and used them the next day. They would work fine. |

| Yes, you can make the SDS-PAGE gels, wrap it in a damp paper towel, label it, place it in a ziploc bag and store at 4C for one week (max). We do it in our lab and it works well. |

| Hello all, The gels can be stored longer then one week. As long as they are stored at 4C in a moist environment, they can be stored for atleast a week. If you wish to store them longer, they can be stored in a small volume (enough to maintain moisture in the ziploc ~3mL) of 0.01% sodium azide solution which will prevent growth. In this condition, the gels can be stored for a few months. |

| U can store it for >2 weeks. I've tried running gels stored for 3 weeks and the protein separation was fine. I usually keep gels (comb attached) in a plastic bag with 1x running buffer, sealed and stored in chiller. |
