It depends on what kind of RNA you are isolating, I mean, is it from a virus' or from the donor cells? Regardless that, I think the only difference between PBMC and plasma should be the RNA yield: i.e. if you are isolating cell's RNA PBMC ought to give you a higher yield.
About the primers, I think there should be no problem as the RNA sequence has to be the same in both starting materials (PBMC and plasma), the only problem should be, as I stated above, the RNA yield as this might affect the detection of the sequence.
