Cool PBMCs total RNA and plasma RNA differences??



Can someone please help with mentioning if thereíre a difference between total RNA isolated from peripheral blood mononuclear cells (PBMCs) and the RNA isolate from plasma?? Also if Iíve a quantification primers and probes for RNA isolated from PBMCs could it be used for the plasma RNA?
This is urgent for me thanks for your help

It depends on what kind of RNA you are isolating, I mean, is it from a virus' or from the donor cells? Regardless that, I think the only difference between PBMC and plasma should be the RNA yield: i.e. if you are isolating cell's RNA PBMC ought to give you a higher yield.

About the primers, I think there should be no problem as the RNA sequence has to be the same in both starting materials (PBMC and plasma), the only problem should be, as I stated above, the RNA yield as this might affect the detection of the sequence.

Thanks a lot for replying, I’m isolating viral RNA. If the only difference is in the yield it won’t be much of a problem for me.

For viral RNA I believe it is best to use plasma in order to avoid unneccesary cell lysing, which will ultimately make the purification step a bit easier, and might improve your RNA yields.
