Default problem in specific mRNA isolation



Hi! Everybody,
I want to isolate my desired mRNA from Taxus leaves, but always what I have are rRNA chloroplastic cDNAs after sequencing. I tried so many times to optimize the RNA isolation protocol and now using RNAesy Mini kit-Qiagen with double amount of PVP. The primers are quite specific. Also the desired cDNA already isolated from Vinca with same primers. Can you give me please any idea?
