Default RNA extraction from Noni leaves



I was extracting RNA from Morinda citrifolia (noni) leaves using CTAB method. This method was practised suitable for RNA extraction from noni leaves.But gel electrophoresis result showed only DNA bands and there is no any RNA bands. I would like to ask does anyone can interpret what may happen to this condition?10s.

Repeat and store very carefully, RNA is more fragile and will degrade before DNA. Make sure you are practicing RNAse free conditions for the extraction. May be a good investment of your time to prepare new buffers, reagents, and autoclave tips etc--and eliminate RNase as a possible variable.

hi

im not sure wheter you have succesfully extract the RNA from noni leaves or not. but im would like to share my expirences here whereby my labmate doing that since last year and she usually use yellowish leaves instead of green young leaves to avoid high polysaccharides and alkaloids that seem higher in green leaves. besides, she using large scale (25ml each reaction) which then washing step done up till 3-4 to remove as much as possible the protein contain.

dont worry since my labmate required 1 year to develop a proper way to extract RNA from by changing the protocol bit by bit. so,try your best
