Hi yfsq,

What is it about miRNA that you would like to discuss?

If I want to transfect to animal cells, which is the better? mature miRNA? or pre-miRNA? why?
Thanks for your answer.

Now I can get myself into trouble, because I remember a story from reading a paper but I don't remember the citation. If anyone has more to add (or to disagree), please jump in!

The unloaded RISC complex (without RNA) associates with the Dicer enzyme. When a pre-miRNA is cleaved by Dicer, the duplex RNA is released in an orientation that allows it easy entry into the RISC complex, where the guide strand is tightly bound and the passenger strand is cleaved by Argonaute. At that stage the RISC complex is ready to test the guide strand against mRNAs.

Based on my recollection of that story, it is the pre-miRNA (the stem-loop hairpin) that would be the most efficient molecule for activating the RISC complex.

I hope that's some help.
