I am doing optimization of RNA extraction from breast tissue. What concerns and precaurions should I take? I got 260/280 ratio ( -3.04) would any body please explain to me what does it mean? first time I am seeing a minus reading.

Some times happened to me also (same breast tissue). I like to know how you prepared RNA, if you using any kit, if yes which one.

After long struggle now am getting 100% good RNA from by combining two kit. First isolate Crude RNA using trizol method (this will eliminate fats from the breast tissue). Then further purify the crude RNA by Quiagen RNA kit (follow RNA clean up protocol in quiagen kit). It always work for me.
