Hey all, I have a question about Real-Time Analysis of Mouse Genotypes. I need to analyze the genotypes of mice that are homozygous and heterozygous. How can I use real-time to assess genotype of the transgenic mice? Does expression always correspond to dosage? Southern blot analysis seems a better approach to me!... tell me please if I may be wrong. Thanks! Ob

You need to know the exact location of the SNP in your sequence. Then design an assay with primers that identify SNP bases. You'll need 2 pairs. One for wild tytpe and the other one for the mutant.

Southern blot is not very accurate and may give false positives

If you need a software that can design SNP genotyping assays, get Beacon Designer or AlleleID. They will help you do [Only registered users see links. ] and design of molecular probes such as TaqMan probes or molecular beacons. These primers can be used for your SNP genotyping and detection Last edited by admin; 04-26-2008 at 08:18 AM.
