Real-Time PCR to detect point-mutation

I have transgenic mice expressing single point mutation. I have few questions:

1. can I use real-time pcr to determine the levels of mRNA of endogenous gene and transgene in Wild Type mouse and transgenic mouse, respectively?

2. I know for WT mouse, i can use general primers (which i can design using beacon software, for eg), can I use primers (18mer) flanking around the point-mutation to do a real-time PCR for tarnsgenic samples??

In other words, can i use region-specific primers to do a real-time pcr?

you may try MGB probe if they are morn than 3 mismatch on probe region
