| Hi everyone! Recently I did a qPCR experiment to measure the relative mRNA levels of dsRNA injected individuals against water injected individuals. A reference gene was used, data were analysed using LinReg. Here is the problem, only 8 biological replicates were used for both dsRNA injected and water control. Variances of both groups are not equal, p value~ 0.00004, data are normally distributed (shapiro) so I asume I would have to use the Welch t-test. However, when I log transform the data, variances are significantly equal p>0.05. Could I do unpaired t-test using log transformed data, or use Welch t-test?
