Beacon Designer 7 is sure the most comprehensive one. Other tools such as Oligo either do not confirm to the qpcr standards or do not have major supporting chemistries.

One thing always to look out for is how well the authoring supports. Going by my experience this vendor (Premier Biosoft) does a pretty decent job. They also offer qpcr assay design services for you if your design is too difficult and still you won't like to shed that much money. This is good if I am designing multiplex PCR assays or alignment assays (cross species or specific to a single sequence).

VectorNTI is ok for planning cloning but for primer and probe design, I personally would never compromise and choose the best.

i think Primer Premier 5 is the best ,but lasergene is fine.
