Question RT PCR Positive Control

Hello everyone,

I just got a question from a fellow user:


I am working with RT PCR. I ran the PCR on the an agarose gel 2%.
The original method uses Real Time PCR (Q-RT) PCR.

However a fellow lab member has changed the method to use traditional RT PCR.

However I tried the lab members primers and program for the machine using the new temperatures for the traditional RT PCR.

The problem is I cannot see amplification of the 700 bp band, I also see amplification of a 150 bp band.

Any ideas?

I think you may be having problems with the primers.

I am not sure, but this seems to be more likely.
