Exclamation Separating yeast from molten agar



Hello,

Does anyone know how I can separate yeast from molten agar? I`m trying to sort out the multiploidy yeast I`ve created using a flow cytometer and I could just make a new culture in broth but I`m worried that the smaller yeast would out compete their larger multiploidy neighbours.

So, does anyone know how I can separate my yeast from my agar? So far, centrifuging and washing are not very effective.

Any suggestions would be greatly appreciated!
