Default how to quantitative detection by sybr green

By real-time PCR with sybr green, sample containing low concentration template can't be detected by first PCR. When first PCR product is as template of nested PCR , sample was positive.
However, how to quantitative detection of original template?

Hmm... tough question. Do you have a known positive control?

You could try dilineating how much is in the original by using a standard and comparing the two.

Either than or try another method of detection.

"dilineating" what means?
I have detected serial dilutions of positive control sample.But after nested PCR, the standard curve relation can't observed .That is serial dilution relation disappear. even very small amount of origin template, the amplification results will be very large after nested PCR.
How to do?
