| Hello. I would like to know how many hours (after anti-CD3 stimulation in vitro) B cells need to change into plasma cells. Last edited by CD19; 06-14-2010 at 09:41 PM. |

| Anti-CD3 soluble or coated on polystyrene plate (dispense 50 µl of 5-10 μg/ml anti-CD3 [OKT3 – Ortho] in sterile PBS incubating for 24 hrs at 4 °C) react with surface membrane T cell receptor (peripheral blood mononuclear suspension in RPMI 1640 medium with 5 % FCS) inducing, after ~16 hrs at 37 °C in 5 % CO2 incubation, cytokines production to achieve stimulus, IL-2 play a major role, on naive B cell changing in B cell blast after ~ further 8 h. From here, time of plasma cell development depending from B-cell subset phenotype (MZ/B1 and B2) with generate plasma blast after respectively 6 and 48 h further. For develop mature plasma cell, is need one progressive down-regulated with rearrangement that to lead at loss of many surface cluster differentiation antigens what it ends in ~24 hrs. The computation time for to complete entire process is ~96 hrs (4 days). |
