i am a new phd student, having problem with the telomerase repeat amplification protocol(TRAP)

i used the milliq water for preparing solutions, n did same as they said but was getting no bands. not even ic band. when i tried higher concentrations of reverse primer,(acx ,once i got five bands but could not repeat it. help me pls
i have been fitin with it since 3 months
