Default S2 Schneider Huge growth



Hello all-

I am culturing S2 Schneider cells. I'm having a hard time controlling cell growth, in that in culture growth goes from 5x10^5 cells/ml (minimum seeding density) to 3x10^7 (maximum reccomended density) in 24 hours.

This is a new cell line in the lab, and no one here has wide experience culturing insect cells. A reccomendation I have seen is to grow them at room temperature, which does indeed slow cell growth by about half but still give the above results. I am going to try 5% HI FBS as opposed to 10% HI FBS today to see if that helps (I viewed this tip on another molecular biology web site).

Does anyone have any insight as to A. why I am seeing such ridiculous growth and B. What I might do to curb cell proliferation other than what I have already tried.

PS there is not evidence of contamination.
