Hello Miss PCR,
mycobacterium tuberculosis is a very interest organism as you probably know. It usually grows inside macrophages, but I am not sure if you will be culturing them like that.

I have used the light cycler before and its one of the fastest real time pcr machines out there (at least several years ago when i used it).

Not sure what you will be doing with syber green I however, its usually used to tag your primer extensions in the real time machine (light cycler) so you can assay gene expression.

If you have any more questions or more specific ones we would be interested in helping.

Good luck
