| Dear western-blot experts, A sales rep. from Millipore just left the lab and almost convinced me to buy what he calls "a revolution in your western-blot experience": a simple device (SNAP i.d.) that makes all incubations shorter by forcing all solutions "through" the membrane (using vacuum). [Only registered users see links. ] It certainly looks like a much simpler, quicker and cleaner way to do all incubation for the westerns, but I would like to hear from people who have actually used the mechine. The question is: will the 850 euros investment increase our happiness? All the best CG |

So far, it has proven to not work as well doing our incubations in little plastic bags. The background is very low (we use 0.125% non-fat milk with the SNAP i.d., and 5% non-fat milk for blots done the old fashioned way), however, the sensitivity is much better the old fashioned way (about 10 - 50 times more sensitive). Since we've gotten our new unit, I've not had any success and I am sticking to my old method until I have more time to troubleshoot it. |

| Yes, this will make your life much easier! It will take a little time to figure out the antibody concentrations for your primary antibody. I use the Snap ID almost all of the time. Some times it will not work with some primary antibodies but I have had a 100% success with all my secondary antibodies that I have used. Additionally it can be incorporated into your traditional WB protocols. Washing steps and blocking steps can be done using the Snap. This is a huge time saver if you have 2 or more blots. I have also determined you can reuse the cassettes but you need to take care of them. I think this thing is the best thing to happen to WB in a long time!!! |
