| I checked some online recipes about how to make sodium ortho-vanadate stock solution, some talk about pH-ing the solution first after the reagents completely dissolve in water, which would then turn to yellow after pH-ing to 10, then the solution needs to be boiled to get rid of this color, this process (pH-ing then boiling) need to be repeated a few cycles until the color is gone after pH to 10. However, some other recipes do not mention this at all. Since quite a significant amout of recipes I checked online emphasize this step, and I really don't want to make a wrong solution for my experiment, I would like to ask if anybody knows why one needs to (or IF one needs to) pH-ing and boiling the solution when preparing sodium ortho-vanadate stock? I came across to a line online saying this would help to activate the chemical compound, however all the websites I checked, including vendor websites who sell this compound, do not mention anything regarding this either. Thanks if you can help me to straight this out! |

| sodium orthvandate : inhibitor of phosphatases~ I just prepare the solution as: Dissolve the sodium orthvandate in ddw (100mM or 200mM), adjust pH to 10 using HCl, the solution may turns yellow,boil the solution until it turns to colorless~repeat the cycle for several times until the solution still colorless and pH stablized at 10. you may store the aliquots at -20c. |

| Hi, Do you guys know how to prepare sodium orthovanadate for inhibition of dynein? Is it the same as mentioned above? Thanks. |
