Default Concentrating adenovirus by ultracentrifugation



Evening all,

Has anyone here concentrated adenoviruses, or any other kind of virus or
similar, by ultracentrifugation? If so, what product did you use, and in
particular, what pore size of membrane?

I'm doing this reasonably successfully using 100 kDa MWCO Amicon Ultras;
however, i get a lot of junk in my retentate, which i assume is serum
proteins, so i'd like to move to a larger pore size, to let that through.
However, i have no idea what molecular mass-based pore sizes mean in
nanometre terms; the various manufacturers' websites are not a lot of use
(notable exception: Pall [1]).

If I were you, I'd actually use ultracentrifugation - viruses are big enough
to pellet while many proteins do (particularly after clarifying filtration
through 0.1 um filters). It's a standard technique with retroviruses and
baculoviruses.
