| I don't know how to find books which about the bacteria mating on agar, so please told me some books or webs to find my question.........thanks a lot~ |

| I don't know a book reference, but the method that has worked best for me is to mix the donor and recipient and pass them through a 0.22 uM filter that's in a screw-apart cartridge. Then you take the filter and put it bacteria side up on an agar plate and grow overnight. |

I just mix both freshly grown donor and receptor strains (sometimes also a helper strain) in 50 ul of NaCl 0.9% and dispense the mix on a 0.22 filter already placed on the agar plate. I incubate the plate face up, overnight, take the filter, wash the cells in NaCl 0.9% and plate an aliquot on the selective media. It works great. Sometimes, when I don't have filters, I just drop the mix directly on the agar. Next day I collect the cells, wash and plate on the selective media. I haven't seen important differences in efficiency... it always works great. Sergio |
