Default Biorad Bradford Assay



Hi OK
Several years ago, we ran into some problems with protein quantitation
for westerns & 2-D work. As I recall - we found that the Bradford
assay was not compatible with detergents. So we switched to another
BioRad assay based on the Lowry method - they call it the "DC" protein
assay. This assay is compatible with detergents. There is also a
modification (treatment with iodoacetamide) for use with samples
containing dithiothreitol or 2-mercaptoethanol.

I am using a Biorad Bradford Protein Assay to estimate total protein

tissue lysates, fir westrn blot analysis. I use a dilution of 1:20 to
1:50
of the lysates to estimate protein. At this dilution I should not get
any
interference in the assay from the SDS, NP40, and Na-Deoxycholate
(present
in the tissue lysis buffer). However the estimation are not always
reproducible.
