| Could it be the Tween-20 concentration. If concentrations of this go up more than the recommended (normally 0.05% for PVDF), I have heard of blots getting such clear backgrounds since they strip proteins from the blots. A drying step before blotting would be a good idea to improve protein binding. I knew of a person in an earlier lab I worked with who used to add T-20 as per his estimation and not using exactly the amount required. he used to have very inconsistent results till we identified his problem. But still a detailed protocol with timings and volumes would help. regards Jayakumar --- |
