Default Sterile lab

Hi
in the near future i will work on animal cell culture, for this we have to establish a lab, that must have a minimum of microbial load, for this please give us some recomendations.
in addition for fuming what are the best chemical that could be used for fumigation in the lab.
is HEPA filter is required with cooling system in the lab or not.

i hope you people will guide me.

hey Afta,
I would only fume initially as its pretty toxic. Its more important to be clean while working to maintain cleanliness.

Some tips on Cell Culturing

1) Always wipe spills of FBS and media in the hood. Bacteria and fungus love this. Use ethanol 70% with distilled water to clean up.

2) We always use Pen. / Strep. Antibiotic and Antimycotic. ie anti-bacterial and anti-fungals in our media. It is VERY helpful in keeping cultures fungus and bacterial free.

3) Once in a while autoclave the cell incubator trays (this could be every few months).

4) Water baths are breeding grounds for bacteria. Always ethanol spray your bottles and wipe them down after water bath incubation!

Formaldehyde vapour can fumigate a lab and cell incubators.

Formaldehyde can be used with potassium permanganate.

Recipe:


* 35 ml formalin (37 or 40% formaldhyde)
* 10 g potassium permanganate / m ^3 of space to sterilize

Mix in a porceline or steel container.

Immediately leave and close the door of the room overnight.


Wear protective clothing, gloves, mask and safety glasses

Avoid inhalation of the fumes

HI the concentration of KMnO4 seems to be high and people say that this reaction is vigorous, needs supervision . how can we leave it overnight? is there any literature support for this.
