Streaking on upper region of 2-D gels

Hello everybody,

my last 2D gels show vertical streakings on the upper region across the whole

gel without affecting lowered parts. I even cannot find help in 2D handbooks.

Hey,

welcome to the forums. a few things you may consider:

1) salt / running / loading buffer (upper buffer on top of gel may be to blame)

2) amount of protein loaded (too high?)

3) 1-D bad?

Just some ideas here.

I ever got that kind of streak too. For my case, I found out that it was because of amount of protein. These clounds appear with coomassie staining but become the spots with silverstaining. Have you ever tried the longer strip?

Hi H,
did you re-cycle your running buffer. If yes, you might want to consider making a fresh buffer.
