Some questions about Liquid Chromatography coupled to Mass Spectrometry...

- how many proteins can they identify?

- what is the cut off for identification? ie how much protein minimum do i need to identify my baby (protein of interest )???

help appreciated....

I did my protein although only one protein of interest.. on the LCMS.. with an ESI.. i was working with about 1 ng amount... not too sure how it would be for your protein. But i think pmoles is a fair bet.. But with a higher amount of proteins present.. i am not sure how it would effect the sensitivity of a single quad ms... why not use a LC-TOF... better sensitivity and mass accuracy..

Hundreds to thousands of proteins can be identified from LC-MS experiments. It depends on many factors such as how long the LC gradient and how much sample you have.
In our lab we can constantly identify lower fmole or even up amole amount of proteins. I believe majority of modern LCMS instruments are capable of doing this .
