Question Streaking of 2-D SDS-Gel Electrophoresis


I am getting streak of my 2-D SDS-Gel.

is it because I am loading too much protein or because the focusing is not working?

Are you getting vertical or horizontal streaking?

Are you loading too much protein?

Have you checked your buffers? for IEF? for the gel?

most likely something wrong with first dimension IEF.
