Unhappy high background

Hi,
My silver staining gels have high background. Can anybody help me on how to reduce the background intensity. I thought it could be because of the water quality but I'm using MiliQ water but still it doesn't help me. Please help me on this. Thank you.

Hi
Why dont you try silver staining step doing at low temperatures(4 C). Use chilled Developer, this can reduce the backgrd intensity..


Do try this and let me know if any improvement

the high background should be related to the short time of incubation with fixation solution, try to leave it overnight in the fixation solution in case of u apply the fisation solution for 30 min or 1h.

Cool Re: high background

I think The high background does not relate to the time of incubation in fixing solution. The background is not different between overnight fixation and 30 min fixation. i used to do both. To ensure about 3 times washing before stain the gel in staining solution. The background will be dark if you wash the gel incompletely.

Do you have a good washing step after you put the silvernitrate on it?
