Default recombinanat protein expression

hi,

i found some problem in expressing my protein in BL21 DE3 host. NO expression in the colony screening. my protein is also non toxic to the host. In the first day iam finding some very minimal expression, next day no expression. IPTG concentration is 1mM. vector i use is pRSETB. kndly guide me in this regard.

very tough observations. Not sure what you could do. Does your vector have high expression (with control protein) in BL21 DE3?

Hi Gajji81,

I also have same problem in the protein expression in BL-21 (DE3). I have used the pRSETA, also no or little expression after IPTG induction. Have you already solved the problem, pls. let me know.
