| Hi I don't know wether or not this is the right place to post but we shall see. I am looking to see if a drug I work with interacts with proteins that my lab has an interest in. Our initial plan was to use size exclsuion chromatography of protein mixed with drug and then measure the fluorescence of the flow-thru, however we found that the drug bound to the column so this ruled out this option. We are now looking to carry out sedimentation equilibrium experiments; spin a mixture of drug and protein of interest and see if the drug cosediments with the protein. The only problem is I have no idea how to carry out any sort of experiment using an ultracentrifuge! Would anyone have any good protocols or advice to get me started? Regards Wandrews |
